Targeting PAQR5 in HCC cells impeded the formation of the immunosuppressive microenvironment in vivo. A-B. the orthotopic liver tumor model in immune-competent C57BL/6 mice validated that knockdown of PAQR5 in mouse cell line: Hepa 1–6 notably inhibited the tumorigenesis and growth. C-E. IHC staining revealed that depletion of PAQR5 in Hepa 1–6 prominently repressed the expression of PD-L1, inhibited infiltration of FOXP3 + Treg cells, while facilitated the CD8 + T cell infiltration. F. WB performed with tumor lysates revealed that the protein level of p-P65 and PD-L1 was significantly reduced in liver tumors with Hepa 1–6 knockdown than in those tumors in shControl group.
